History of 4-AcO-DMT

What is 4-AcO-DMT?

4-Acetoxy-N,N-dimethyltryptamine (psilacetin, O-Acetylpsilocin, or 4-AcO-DMT) is a synthetic tryptamine with psychedelic properties which are closely related to active compound found in magic muchrooms, psilocybin and psilocin. It bears a striking molecular resemblence to these psychoactive molecules.

Psilocin was extracted, isolated, and acetylated by Dr. Albert Hofmann in the 60s along with Franz Troxler, for Sandoz Ltd. The company also holds the patent for 4-AcO-DMT. Due to the acetylation of psilocin, 4-AcO-DMT is also called 4-Acetoxy-DMT as well.¹

Dr. Hofmann acetylated the psilocin molecule, presumably in order for longer-term storage of psilocin. Psilocin may lose potency over time, however, Acetylated Psilocin in fumarate or HCL form don’t lose their efficacy. Another reason for acetylation of the molecule was due to how acetylated drugs deacetylate and affect the metabolism of the drug.²

Acetylation is a chemical reaction which introduces an acetyl functional group into a chemical compound. This process results in what is known as an “Acetoxy” group. This is a substitution of an acetyl group for an active hydrogen atom. Acetoxy group, shortened to AcO or O-Ac deacetylates upon first pass metabolism in the body.³

Many drugs are acetylated to study their deacetylation effect on the body. In fact, a multitude of drugs and various other compounds require acetylation for effective metabolism. 4-AcO-DMT is the acetylated form of 4-HO-DMT (Psilocin) which itself is a metabolite of Psilocybin Mushrooms.⁴

It can said to be a pro-drug for psilocin. 1 Can you use 4-AcO-DMT as a substitute for mushrooms?

Yes. It is safe and effective. It is a functional analog of pilocybin mushrooms, and chemically, and effecively features the same properties. People are using 4-AcO-DMT to microdose more accurately than with mushrooms.

4-AcO-DMT Is Either Not Illegal Or Unscheduled In The Following Countries:

• Germany (Illegal under Analog Act as it is an Ester of Psilocin)

• United States: 4-AcO-DMT is unscheduled in the United States, however, 4-AcO-DMT is considered a Research Chemical or a designer drug sold by many online vendors. 4-AcO-DMT is an analog of psilocin and as such legal for labratory research. Possession of 4-AcO-DMT for the purposes of the sale for human consumption can be illegal under the Federal Analogue Act, under which any substance “substantially similar” to an illegal substance listed in Schedule I or II is treated like those in schedules, but only if intended for human consumption.

Pro Drug Theory

Upon first pass metabolism, the Acetyl (acetoxy) group is metabolized into a hydroxel group, effectively turning 4-AcO-DMT into 4-HO-DMT which is the chemical name for Psilocin.⁵

During the first pass metabolism and additional passes through the liver, 4-AcO-DMT is broken down or deacetylated into Psilocin. The Acetoxy group is broken off and only a hydroxyl remains on the fourth position. This rapid deacetylation to Psilocin is why 4-AcO-DMT hits faster and harder than mushrooms.⁶⁷

The same thing happens with Psilocybin which is 4-PO-DMT. Upon first pass metabolism, the PO (phosphoryloxy) group loses the phosphate, and turns into a hydroxel group, turning 4-PO-DMT into 4-HO-DMT.We can say that 4-PO-DMT (Psilocybin) and 4-AcO-DMT (Acetylpsilocin or 4-Acetoxy-DMT) are pro-drugs for Psilocin (4-HO-DMT).

The prodrug theory of 4-AcO-DMT has been criticized, however. The very fast onset of the psychoactivity of 4-AcO-DMT when intravenously injected or vaporized, bypassing the first pass metabolic function, suggests the compound is fairly psychoactive on its own and not a metabolic pro-drug or molecular pre-cursor to psilocin.⁸

However, during my research of acetylated molecules, I found that the deacetylation is what causes the rapid onset.

Subjective difference have been noted by trip reports presented on Erowid.⁹¹⁰ Some report the effects of 4-AcO-DMT to be identical to those produced by psilocybin mushrooms, while others note differences. Some people find it more mellow, more easy, or less harsh than mushrooms. Some have even described as a level 1 DMT trip that lasts for hours, particularly at high doses.

Even though the substance dates back to the 1960s, it has a limited history of human consumption. It wasn’t until a decade or so ago that 4-AcO-DMT came to light as a Research Chemical and as a pro-drug for Psilocin.

Pharmacology

4-AcO-DMT has the same basic molecular structure as both psilocin and psilocybin, but with an acetoxy functional group (AcO) attached to its indole ring. The chemical forumula for Psilacetin is C14H18N2O2. ¹¹

We can safely assume, given the chemical and functional similarities to psilocin and psilocybin, that 4-AcO-DMT has roughly the same mechanism of action used to activate the 5HT receptor agonism.

4-AcO-DMT is primarily a serotonergic agonist, and it’s psychoactivity comes from the binding activity at 5HT1 A, B, C, and 5HT2 receptors. The reason why 4-AcO-DMT also helps with migraine headaches is because of its efficacy at narrowing blood vessels by targeting the 5HT1D receptors known for its vasocontriction effects.

There are two important effects that come from serotonin agonist caused by 4-AcO-DMT:

1. It produces what is called Brain Derived Neurotrophic Factor (BDNF). BDNF is “like Miracle-Gro for your brain. It stimulates growth, connections, and activity.”

2. The heightened levels of Glutamate transmission in the brain. Glutamate is the neurotransmitter which is most responsible for cognitive, learning, and memory related brain functions.

Invetigations into Glutamate and BDNF functionality and partnership in the brain are still under research, though it has been shown that higher amounts of the two will result in extremely positiv results.¹²¹³

Psychedelics work as liasons for these connective functions by slightly mitigating the activity the “Default Mode Network” (DMN). In mental activities such as daydreaming, self-reflection, and temporal thougth processes, such as thinking about the past or future, high amounts of DMN activity has been reported. ¹⁴

Studies have suggested that depression may be caused by an abundance of DMN activity, resulting in self-desctrive thoughts, repetition over analysis, and self-criticism, which are all symptoms of depression. ¹⁵

Perhaps this is why psychedelic such as LSD, mushrooms, 4-AcO-DMT etc. are so effective for depression and anxiety. They inhibit the DMN funcitonality to allow the person to be in the present moment and focus on what is at hand, instead of being plagued by thoughts of the past, or worries of the future.

Another great benefit, shown through scientific research, is that it helps to treat depression. A single administration of the hallucinogen, 4-acetoxy-dimethyltryptamine, prevents the shift to a drug-dependent state and the expression of withdrawal aversions in rodents¹⁶

Effective Dose

What is a safe dosage of 4-AcO-DMT?

4-AcO-DMT is rather comparable to magic mushrooms, and the dosage amounts are very similar.

Duration

Many people have reported little difference between 4-AcO-DMT and Psilocybin Mushrooms other than the rapid onset. Where mushrooms are a gradual build up over an hour, O-Acetylpsilocin takes roughly 15-20 minutes.

As mentioned previously, 4-AcO-DMT hits faster and harder due to it being an Acetylated molecule. Deacetylation happens faster than first pass metabolism.⁷

The trip length is somewhat proportional to how much you ingest.¹⁷

Toxicity and LD-50

The Toxicity and LD-50 data on 4-AcO-DMT is lacking due to it’s lack of popularity and a limited history of human use. However, since it is a prodrug for Psilocin, it is very similar to the LD-50 of mushrooms. So far no deaths have been reported from this substance.

Those who have tried 4-AcO-DMT have yet to report any negative health issues associated with its use. I have personally eaten, drank, smoked, and vaporized 4-AcO-DMT in large amounts, even mixing it with large doses of Harmala, DMT, LSD, 25i-NBOME, Ketamine, Amanita Muscaria, Salvia Divinorum and many others, and I can report that it’s rather harmless when done within reason and balance.

Always start with low doses. You can always take more, later on. Harm reduction is about being conscious, aware and personally responsible.

Safety

4-AcO-DMT is extremely safe to use and microdose as it is the acetylated version of psilocin. The only thing one has to worry about is overdosing. It is easy to overdose on 4-AcO-DMT than it is on magic mushrooms. It is one of the safest 4-substituted Tryptamines next to psilocybin and psilocin.

It’s basically just Psilocin with an Acetoxy functional group bonded to the indole ring, instead of a hydroxy, and it acts as a pro-drug for psilocin.

There are a few potential problems with microdosing 4-AcO-DMT, though. Higher doses such as 150mg or more can cause vomiting or diarreah, and muscle spams.

I have personally taken 280mg of 4-AcO-DMT a few times, combined with Hamrla alkaloids, and speaking from personal experience, it’s just a little harsher on the body, but stil completely safe.

The highest concentrations of Serotonin are located in the gastrointestical tract. The more serotonin agonism there is, the higher the chances of expelling the gut, so just make sure to keep a light fast, and keep your stomach empty before taking 4-AcO-DMT in higher doses.

When I vaporized or smoked 4-AcO-DMT with Cannabis, the onset was within 1-2 minutes, and 10 minutes later I felt muscle spams. I was shaking, strongly. Though it was not painful, it was a bit uncomfortable. I am prone to be colder than most other people, so perhaps it could have simply increased my sensitivity to temperature.

There have not been many reports of adverse reactions to 4-AcO-DMT.

Sometimes if you take a higher dose, it will be a slightly more difficult trip, but nothing to worry about. So long as you have a well calibrated milligram scale, and you measure out a dose properly, up to 75mg will be an easy trip.

Cardiotoxicity

The safety properties of of 4-AcO-DMT have not been studied in great detail due to it being a novel and rather less known tryptamine, however, its chemical and functional similarities to psilocybin and psilocin seem to suggest that the potential risks are likely comparable to their Hydroxy and Phosphoryloxy relatives.

4-AcO-DMT is physically not habit-forming. It also does not cause addiction. The tolerace is built up rapidly, like mushrooms or LSD, and eventually even extremely high doses stop working (upon repeated use). With very high does, there is, however, the potential for heart problems or cardiotoxicity.

The highest concentrations of serotonin are in the gut. The next highest concentrations of serotonin is in the muscular system. The heart is a muscle that pumps blood. Due to the fact that 4-AcO-DMT, like psilocin, targtes the 5HT1D receptor, it causes tightness of the muscles due to its vasocontriction properties.

As I mentioned before, I have personally taken 280mg of 4-AcO-DMT many times, and I’ve never had any heart issues. Though, everyone is different, and it may hold a risk potential at higher doses.

A standard dose of 4-AcO-DMT which is 25mg will most likely not carry any cardiotoxicity at all. Even 50mg (equivalent to 5 dry grams of psilocybin, a heroic dose) will be just fine.

In some rare cases, if one has a weak heart muscle or any other cardiovasular issues, 4-AcO-DMT may cause some cardiotoxicity and and a general painful feeling in the chest, but that vanishes in a few days, and it’s not a long lasting effect. ¹⁸

Just make sure to not take extremely high doses of 4-AcO-DMT and you’ll be fine.

Is 4-AcO-DMT safe to use with other drugs?

Drug interactions are always something to watch out for. It is important to experience the drug for itself first, and to familiarize yourself with its effects, both physiological and psychological before attempting to mix it with something else.

4-AcO-DMT’s Dangerous Drug Interactions

• Alcohol

• Antihypertensives (blood pressure medications)

• Antipsychotics

• Any CNS (Central Nervous System) depressants (Xanax, Ativan, etc)

• Any drug containing dextromethorphan/DXM.

• Lithium

• Tramadol

• Barbiturates

• SSRIs or SSNRIs (avoid DMT if on anti-depressent medication)

• Vasodilators

(I made sure to simply avoid these and never bother. I don’t enjoy any of these drugs or the effects and feelings that they produce. I’ve never found any of these drugs to add anything to the experience or to my consciousness. I only like psychedelics.)

• 25i-NBOME (As long as no MAOi was consumed, it is a wonderful combination. Euphoric and extremely introspective)

• 2CB (As long as no MAOi was consumed, same as 25i, but more physically pleasing and sexual)

• Amanita Muscaria (Amazing space. The dissocative factor coupled with 4-AcO-DMT’s center stage presentation is very illuminating.)

• Cannabis (My staple. I consume cannabis everytime I take psychedelics.)

• Ketamine (What a beautiful combination. The dissocation coupled with how 4-AcO-DMT puts you at the center of the stage, and to detached from the concept of the “self” is phenomenal.)

• LSD (Smoking Harmala and/or 4-AcO-DMT on the peak of an acid trip is gorgeous. It sends the LSD into a frenzy. Not recommended for beginners.)

• MDMA (As long as no MAOi was consumed. This is one of the most amazing combinations I’ve ever tried.)

• Mescaline (As long as no MAOi was consumed. Very similar to 25i’s experience)

• Psilocybin

• Salvia Divinorum I’ve combined a Salvia 20x Level 5 hit with 75mg 4-AcO-DMT fumarate in a bong, and then continued to smoke Salvia all through the night. Read about the most amazing and therapeutic trip on Salvia

• Scopolamine (Not as much fun as I thought. Scopolamine is a dangerous substance, like Dutura, and it’s absolutely not recommended unless you know what you are doing. I’m very serious.)

4-AcO-DMT Effects

What are the physical and psychological effects of 4-AcO-DMT?

The effects of 4-AcO-DMT are almost identical to Psilocybin. Since it is acetylated, it hits you faster and stronger than psilocybin. Mushrooms can take up to an hour and a half to take effect, whereas Psilacetin starts taking effect in 15-20 minutes after ingestion.

Many people have reported that 4-AcO-DMT begins in a way similar to how DMT begins. Many people report that it’s a much cleaner trip. The psychedelic community is seriously starting to consider it as a safe alternative to psilocybin mushrooms. ¹⁹

Visual Effects:

Enhancements: 4-AcO-DMT works like psilocybin for visual enhancements which include:²⁰

1. Enhanced visual acuity. You notice things, even slight movements farther away.

2. Colors become brighter and more distinquishable.

3. Edges are sharpened, which helps in movement detection.

4. Depth perception is greatly improved. It almost feels like one can zoom in with one’s eyes.

5. Greatly enhanced pattern recognition, especially in shag carpeting or sand.

Distortions: Ther are visual distortions and modifications to vision. The following distortion effects are produced:²¹

1. Drifting. Morphing. Melting. This is a highly pronouced effect, specific to DMT in my personal experiences.

2. There are always tracers, which are fun to watch.

3. You also see after-images when you look at something else or shift your gaze.

4. Repetition of textures, and the slight appearance of bump-mapping (a 3D rendering technique used on computer science).

5. Shifting and blending colors.

6. Slicing. It not only feels like, but seems like a sharpt jagged edge is slicing in your your mind or your thoughts, and cutting things evenly, sometimes painfully.

Geometric Effects:²²

1. There are infinitely regressing patterns, especially in darkness.

2. Patterns are circular in nature.

3. There is a certain geometric fractal pattern which repeats itself as you go ever deeper into the vision.

Auditory Effects

1. Enhancements: hearing ability is heightened, and more sentivity to noise. That can be a blessing or a curse. Usually I find that if my environment is not quiet, I am severely bothered by any noise, or my increases sentivity to pick up noise.

2. Distortions: Sounds can often blend into each other, and sometimes there is a peculiar sound as if a crumbling of paper or plastic cellophane wrapper.

3. Hallucinations: You can definitely hear sounds which may not be there, or might just be in your own head, though this does not happen often.

Physiological Effects

One can expect the same effects from 4-AcO-DMT as from psilocybin mushroom due to their molecular and functional similarity. However, there are two primary effects I have noticed that are unique to 4-AcO-DMT:²³

1. Extreme internal weather changes, ranging from a feeling of hypothermia to being really warm. Muscles spasm occur in my calf muscles and I am extremely uncomfortable for 15-20 minutes. This effect only happens when I vaporize 4-AcO-DMT. This effect does not happen when I eat or drink 4-AcO-DMT.

2. Extreme Sedation and Yawning. No matter if I smoke, vaporize or eat 4-AcO-DMT, depending on dosage level I go through extreme yawning and feel very sedated and sleepy. The most I’ve ever taken at once time was 280mg of 4-AcO-DMT and I yawned hard for about an hour.

Other than these two effects, the rest is fairly identical to psilocybin. Though, a lot of people have also said that psilocin makes you yawn a lot.

Psychological Effects

The psychological experience of 4-AcO-DMT is similar to Psilocybin Mushrooms apart from a few key things that I noticed. The most prominent of psychological effects generally include:²⁴

1. Conceptual thought processes amplification.

2. Connectivity of individual thought patterns and pattern recognition.

3. Profound feelings of Deja-Vu, that this existence has been lived before.

4. Certain delusions are produced, such as being God, or a Prophet with revelations you feel compelled to share with others.

5. Direct Communication with the subconsious, the higher self, or God (Divinity).

6. Total loss of the I, the Individual, the “Self”. Profound ego suppression, loss and cultural death.

7. Enhancement and Depressive Waves: Described as constant waves of extreme sense and thought stimulation. Profound thought patterns that are spontaneously. In a cyclic fashion, like waves, thoughts and understanding of those thoughts flows in and out. The two peculiar mania and depressive states seem to alternate in a consistent loop once every 10-15 minutes.

8. Enhancement of creativity, reasoning ability, and generally held current mind state.

9. Enhanced feelings of importance, fascination with everything, and being awe-struck.

10. Coincidentia Oppositorum, the meeting of opposites feelings without clash.

11. Feelings of total free will coupled with determinism.

12. Intropsection, Outrospection, and Retrospection.

13. Destruction of the cultural thought processes and the removal of any cultural filter, allowing direct access to the being that is the human underneath.

14. Profound states of unity with mind, body, and soul. Recognition of the interconnectedness between all molecules as if Quantum Physics just made sense in totality.

15. Time dilation, a sense of gravity modification, like Salvia Divinorum, and slight time distortion.

4-AcO-DMT is dark. Dark in the sense that it forces you to look at the darker parts of your psyche more so than mushrooms. After having consumed 10+ grams of 4-AcO-DMT over the last few years, compared to hundreds of mushroom journeys I’ve taken, I’ve noticed a difference in the mood.²²²³

4-AcO-DMT, to myself, seems to be more spiritual in nature. Though, it really made me question what spirituality is. Is there such a thing as spirituality? I mean, the holy man comes and sells me the concept of God. The psychologist sells me the ego and the self. The spiritual guru sells me spirituality and Nirvana. Does anyone have an idea of what’s going on or is even spirituality a cultural concept? Could it be that there is a spiritual culture? And as I’ve discoverd, any culture is the enemy of the individual.

4-AcO-DMT has a unique … overtone… to the experience. Different strains of mushrooms seem to produce similar psychological experiences which are very slightly qualitatively different from each other.

For example, some people love Cubensis, and say that Golden Teacher or Golden Cap offers a slightly different experience than Mexicana etc. This is similar to how the same strain of Cannabis can affect two people completely differently.

How To Microdose 4-AcO-DMT

According to Erowid²⁵, magic shrooms are roughly 0.5% psilocin by weight, so about 5mg per gram of dried shrooms. Psilocybin mushrooms contain 0.63% psilocybin and 0.60% psilocin. Therefore, 1-gram of shrooms has 6 mg each of psilocin and psilocybin. The psilocybin dephosphorylates to roughly 4.5mg of psilocin, which totals to approx 10mg of psilocin per gram of dried mushrooms.²⁶

Calculating 4-AcO-DMT Microdose

The molar mass of 4-HO-DMT, or Psilocin is around 204.273 g/mol. Psilocybin’s molar mass is 284.25 g/mol.²⁷

4-AcO-DMT’s molar mass is about 246.3049 g/mol for freebase, and a molar mass of 116.07 g/mol for fumaric acid which adds to a total of 362 g/mol. 4-AcO-DMT is sold as fumarate salt or hydrochloride (HCL). The molar masses are 4-AcO-DMT HCl - 282.7649 g/mol, and 4-AcO-DMT Fumarate - 304.3399 g/mol respectively.²⁸²⁹

(362.4 / 204.3) * 5 = 8.869. We’ll round it off to 10. One gram of dried psilocybin mushrooms is roughly equal to about 10mg 4-AcO-DMT, for simplicity’s sake.

Microdosing Methods

Without a scale that is able to measure in milligrams, 4-AcO-DMT can be very tricky to microdose. It is recommended that you get a scale, otherwise please do not attempt to consume 4-AcO-DMT, as eyeballing or miscalculating can turn out to be potentially dangerous. For harm reduction purposes, it is essential that you get a scale that can measure to the milligram, that is, to 0.001g.

If you don't have a milligram scale, do not attempt to consume 4-AcO-DMT fumarate or any powder form.

Accurate dosages can only be measures using an electronic milligram scale. If your measurement is off by even a few milligrams, the microdosing effects would be negated. You would either not take enough, or take too much.

Volumetric Dosing

Psychonaut Wiki’s Volumetric Dosing Guide is one of the best methods by which to consume microdoses of 4-AcO-DMT, among many other substances.

Here’s how to prepare 4-AcO-DMT microdosing solution in a few easy steps using volumetric dosing. You will need the following supplies.

• Get an Electronic Milligram Scale

• Get Everclear, Vodka, or >40% Alcohol

• Get 100ml syringes or droppers.

• Get a 100ml or 200ml vial or dropper bottle.

• Drop 10mg of 4-AcO-DMT into the 200ml vial/bottle. Measure out 100ml of alcohol in the syringe. Put 100ml of alcohol in the bottle containing the 10mg of 4-AcO-DMT. Let this fumarate salt powder dissolve overnight.

• Close the bottle with the cap, and shake it vigorously to completely dissolve the substance into the solution.

• Using the syringe to measure out 25ml of the solution, which now contains 2.5mg of 4-AcO-DMT. 25ml is 1/4th of 100ml, that is, 2.5mg is 1/4th of 10mg of 4-AcO-DMT.

Ethanol Solution

To prepare 4-AcO-DMT for microdosing, start by measuring out 10mg on an electronic milligram scale. Add this to 100ml of Everclear Ethanol. Now, 25ml of this solution will contain 2.5mg of 4-AcO-DMT.

Please keep in mind that Ethanol is the best for mixing 4-AcO-DMT. Psilacetin oxydizes in water rather quicky for it to be a long term storage solution. Everclear or Vodka are the best option for psychedelic tryptamines, especially 4-substituted tryptamines, as they will oxidize rather quickly in distilled water.

Alcohol also ensures the solution remains free of any mold or bacterial growth. Many people like to use Everclear, Vodka, but almost any non-flavored alcohol solution which is greater than >20% would be fine for sterility and long term solution.

If you don’t have a milligram scale, volumetric dosing will not work, you will not be able to measure a proper dose.

Capsules

A lot of people don’t like the bitter taste, so they order empty capsules from Amazon and fill them up with the measured dose. It can be a bit tedious to individually measure out 2.5-mg, but some people don’t seem to mind the effort.

If you don't have an accurate milligram scale, I don't recommend consuming 4-AcO-DMT. Period.

Mixing in Food & Drinks

After having measured out the correct dosage, feel free to consume it with food or drinks per your taste by mixing in 25ml of the solution you prepared previously using 10mg of 4-AcO-DMT and 100ml distilled water or ethanol.

Apart from 4-AcO-DMT’s dose difference, almost everything mentioned in How to Prepare a Psilocybin Mushroom Microdose and How to Microdose Acid and Magic Mushrooms? is applicable. Follow those guides and stay safe.

Blotter Tabs

If you have measured blotter tabs of 4-AcO-DMT, start with finding out exactly what-mg they are. Usually, blotter tabs come in 20-mg - 25-mg. If that’s the case, and you have, for example, 20-mg blotter tabs of 4-AcO-DMT, then start by cutting the 20-mg tab into two equal parts to get 10-mg halves. Now divide these in half again to get four 5-mg squares. Now cut each 5-mg square in half to get 2.5-mg pieces. A microdose of 4-AcO-DMT is 2.5-mg.

In fact, after having microdosed on over a dozen 4-substituted tryptamines, I have found that almost all psychedelic tryptamines (with the notable exception of DMT) function in “almost” exactly the same manner in tiny doses such as microdoses.

Essentially, microdosing LSD or Psilocybin or 4-AcO-DMT (and many other psychedelic tryptamines drugs) in micro doses function and behave in exactly the same manner. Please keep in mind that Ethanol is the best for mixing 4-AcO-DMT. Psilacetin oxydizes in water rather quicky for it to be a long term storage solution. Everclear or Vodka are the best option for psychedelic tryptamines, especially 4-substituted tryptamines, as they will oxidize rather quickly in distilled water.

Alcohol also ensures the solution remains free of any mold or bacterial growth. Many people like to use Everclear, Vodka, but almost any non-flavored alcohol solution which is greater than >20% would be fine for sterility and long term solution.

If you don’t have a milligram scale, volumetric dosing will not work, you will not be able to measure a proper dose.

Tolerance to 4-AcO-DMT

4-AcO-DMT produces a tolerance similar to LSD, which lasts up to two weeks, after which the tolerance resets back to baseline. It also similar to the tolerance produced by magic mushrooms and any other tryptamines.

In short, 4-AcO-DMT mimimal tolerance lasts up to a week, and to return to baseline takes two weeks. The longer the time between trips, the stronger the trip will be.

This does not mean that you can continue to take higher and higher amounts, as you’ll just end up torturing your serotonin receptors, and since you’ll be putting in too much serotonin in the brain, the brain itself will think, “Gee, there is already a surplus of sertonin, so I should stop making my own” resulting in extreme depression once you stop dosing 4-AcO-DMT.

Tolerance is rapidly built, immediately. From personal experimentation:

Day 1: 50mg of 4-AcO-DMT feels like 5 dry grams of mushroms. Day 2: 50m of 4-AcO-DMT feels like 2.5 grams of dry mushrooms. Day 3: 50mg of 4-AcO-DMT feels like 1.5 grams of mushrooms. Day 4: 50mg of 4-AcO-DMT feels like 0.5 grams of dry pislocybin mushooms. Day 5: 50mg cannot even be noticed. It basically feels like a microdose.

It also has a cross tolerance with anything else that effects the 5HT receptors, such as LSD, magic mushrooms, and other 4-substituted tryptamines.

It does not however, have any tolernace relation with N, N-DMT.

Comparison to Psilocybin

How does it compare to Psilocybin Mushrooms?

Due to the acetylated functional group, it metabolizes faster than the dephosphorylation of Psilocybin. Where Psilocybin takes up to an hour and a half to metabolize the phosphoryloxy group (4-PO-DMT) into a Hydroxyl, the deacetylation of psilacetin is much faster.

Deacetylation of 4-AcO-DMT rapidly convert the Acetoxy group to a Hydroxyl (HO) in under 20 minutes. 4-AcO-DMT hits you harder and faster than mushrooms, however, the body load is slightly less that of Psilocin.³¹

What’s important to note here that the there is a great variation in the naturally occurring levels of psilocybin and psilocin even between the same species of mushrooms, and no two experiences are likely to be the same, though similar. The trip is also affected by other alkaloids which are present in the mushrooms naturlly, such as baeocystin and norbaeocystin.³²

Comparison to DMT

How does 4-AcO-DMT compare to DMT?

4-AcO-DMT is pure Acetylated Psilocin. It does not contain Baeocystine or Norbaeocystine as well as other alkaloids found in mushrooms. It begins similarly to how DMT begins.

You can rapidly tell the difference after the first minute in how clean this feels. It’s much less of a body load than DMT. On DMT the body is hammered hard instantly, but that’s not the case with 4-AcO-DMT.

Vaporizing 4-AcO-DMT

Vaporizing 4-AcO-DMT begins almost exactly like DMT and starts to become different after about 1-2 minutes.

A lot of people even say that 4-AcO-DMT is like a level 1 (weak) DMT trip that lasts several hours.

Mindful & Conscious Microdosing

I find it essential to keep a daily journal of my activities, feelings, emotions, and thoughts. Be mindful and aware] as you observe your daily life. Read about how to prepare a psilocybin mushroom microdose.

Observe your thoughts. Be attentive to your mind and body. Observe any changes in your desire for various foods or pleasures. What do you find yourself wanting to do? I’ve observed that microdosing psychedelic tryptamines give me a clarity of perception with which I can observe the complete interaction of this living and thriving organism in the Universe in which it dwells.

Conclusion

4-AcO-DMT is almost identical to Psilocybin. It’s called “mushroom powder by many, including myself. It’s consisent in dosage, and doesn’t vary in potency as pislocybin mushroom do. It’s safe to use as a mushroom alternative and I find it to be a lot more spiritual and clean than mushrooms.

It’s definitely an alternative for those who dislike mushrooms, but want the full mushroom experience. Remember to be safe out there and practice Harm Reduction though knowledge, information gathering, and by being aware of what you’re doing.

Even though the experience of 4-AcO-DMT can never be the same as “shrooms” due to the lack of many other alkaloids found in mushrooms, it’s almost identical in effect. Some might say it’s cleaner. 4-AcO-DMT hits you harder and faster than mushrooms do.
